Antisera for tissue-typing: the production of cytotoxic and agglutinating antibodies following intradermal leukocyte injections in man.

چکیده
L EUKOCYTE TYPING may prove to be an important guide in determining histocompatibility relationships between donor and recipient in tissue l 2 Most studies directed towards ascertaining correlations between human leukocyte factors are carried out with batteries of multispecific antisera. The reactions of such antisera tend, unfortunately, to mask antigenic differences between closely linked factors.3 Monospecific antisera are needed for the reliable delineation of these factors, as well as for genetic studies of histocompatibility antigens.4’5 The present work was undertaken in an attempt to produce monospecffic antisera by immunization of recipients with leukocytes from closely matched partners.

Advantages of Placenta as a Source for Mass Production of HLA-Antisera
Human Leukocyte Antigens (HLA) of fetus are responsible for induction of HLA antibodies in mother. Exposure to the mismatched paternal HLA antigens in the fetus gives rise to a polyclonal antibody response against paternal HLA antigens. HLA antibodies are present in the maternal circulation of the placenta and are absorbed onto the placenta [1]. The major source of HLA antisera is from sera of ...

Background: Fasciolosis is a worldwide disease with major economic and public health consequences. Early detection of the infection is important for the prevention and control of the disease. ELISA allows for early detection of fasciolosis in man and animals. Fasciolosis is caused by Fasciola hepatica and F. gigantica in man and domestic animals respectively. These two species have many similar...
متن کاملAllogeneic major histocompatibility complex‐mismatched equine bone marrow‐derived mesenchymal stem cells are targeted for death by cytotoxic anti‐major histocompatibility complex antibodies
BACKGROUND Allogeneic mesenchymal stem cells (MSCs) are a promising cell source for treating musculoskeletal injuries in horses. Controversy exists, however, over whether major histocompatibility complex (MHC)-mismatched MSCs are recognised by the recipient immune system and targeted for death by a cytotoxic antibody response. OBJECTIVES To determine if cytotoxic anti-MHC antibodies generated...
متن کاملTHE PRODUCTION OF MURINE MONOCLONAL ANTIBODIES (MAb) DIRECTED AGAINST HUMAN T- LYMPHOCYTE SUBSETS
The production of murine monoclonal antibody (MAb) has not yet been reported in Iran. The present work describes for the first time the generation of several murine hybridoma clones secreting MAbs directed against human leukocyte surface antigens. The secreted antibodies by hybridoma clones have been screened on different lymphoid and non-lymphoid tissues. Results indicated that of seven h...
